Maintaining genome integrity.

چکیده

Introduction Genomes have always been vulnerable to damage from intraand extracellular sources such as oxidative species generated by cell metabolism, spontaneous base loss or modification, radiation and chemicals (Friedberg et al, 2005a). As a result, mechanisms to cope with DNA damage developed early in evolution and have been conserved. Organisms need some genome stability, but must allow mutational changes to drive adaptation and evolution. How this is achieved has been the subject of much interest primarily because, in humans, defects in maintaining genome stability are associated with susceptibility to cancer, abnormal immune responses and genetic disease (Friedberg et al, 2005a). This conference on Responses to DNA Damage conveyed an understanding of the complexity of repair and included reports on the intimate details of how enzymes recognize and remove certain DNA lesions. Fig 1 summarizes the DNA repair mechanisms covered in this report; it shows those that remove DNA damage and those that allow cells to tolerate damage.

Blockade of Pachytene piRNA Biogenesis Reveals a Novel Requirement for Maintaining Post-Meiotic Germline Genome Integrity

Piwi-interacting RNAs are a diverse class of small non-coding RNAs implicated in the silencing of transposable elements and the safeguarding of genome integrity. In mammals, male germ cells express two genetically and developmentally distinct populations of piRNAs at the pre-pachytene and pachytene stages of meiosis, respectively. Pre-pachytene piRNAs are mostly derived from retrotransposons an...

Aicardi–Goutières syndrome protein TREX1 suppresses L1 and maintains genome integrity through exonuclease-independent ORF1p depletion

Maintaining genome integrity is important for cells and damaged DNA triggers autoimmunity. Previous studies have reported that Three-prime repair exonuclease 1(TREX1), an endogenous DNA exonuclease, prevents immune activation by depleting damaged DNA, thus preventing the development of certain autoimmune diseases. Consistently, mutations in TREX1 are linked with autoimmune diseases such as syst...

Archaeal Genome Guardians Give Insights into Eukaryotic DNA Replication and Damage Response Proteins

As the third domain of life, archaea, like the eukarya and bacteria, must have robust DNA replication and repair complexes to ensure genome fidelity. Archaea moreover display a breadth of unique habitats and characteristics, and structural biologists increasingly appreciate these features. As archaea include extremophiles that can withstand diverse environmental stresses, they provide fundament...

The MCM helicase: linking checkpoints to the replication fork.

The MCM (minichromosome maintenance) complex is a helicase which is essential for DNA replication. Recent results suggest that the MCM helicase is important for replication fork integrity, and may function as a target of the replication checkpoint. Interactions between MCM proteins, checkpoint kinases, and repair and recovery proteins suggest that MCMs are proximal effectors of replication fork...

Break-induced DNA replication.

Recombination-dependent DNA replication, often called break-induced replication (BIR), was initially invoked to explain recombination events in bacteriophage but it has recently been recognized as a fundamentally important mechanism to repair double-strand chromosome breaks in eukaryotes. This mechanism appears to be critically important in the restarting of stalled and broken replication forks...
